Postdoctorate from Department of Gastroenterology Publishes Article on Stromal Cells Immune Training Mechanism in Top Immunology Journal Science Immunology

Recently, Physician Wu Dongwen, postdoctorate candidate of Department of Gastroenterology of Xiangya Hospital published original article titled “IL-17–dependent fibroblastic reticular cell training boosts tissue protective mucosal immunity through IL-10–producing B cells” inScience Immunology(IF: 17.73, Q1), revealing new mechanism of immune training. Dr. Wu Dongwen is the independent first author, Department of Gastroenterology of Xiangya Hospital, Central South University is the first affiliation.

Nonspecific immune responses to pathogens are heightened in individuals previously exposed to pathogen-associated stimuli, a phenomenon known as Immune Training, which is fundamentally accomplished through metabolic reprogramming and epigenetic changes. The study first elucidated on the necessity of IL-17 signaling in fibroblastic reticular cells (FRCs) in the development of protective antibody responses and prevention against pathogen reinfection. It then validated that the FRC metabolism and epigenetic changes induced by theDextran Sulfate Sodium Salt(DSS) enteritis significantly enhanced the antibody response of mice to Citrobacter Rodentium and that IL-17 is of key importance in the inflammatory “training”. Finally, through experiments on FRC specific IL-17Ra knocked-out mice and B cell specific IL-10 knocked-out mice, the study revealed that FRC-intrinsic IL-17 signaling and the IL-10 production by activated B cells were required for colon protective effects of inflammatory training. This study not only validated the key importance of FRC activation in lymphatic drainage to protective humoral immunity, but also provided new perspectives for understanding intestinal mucosal immunity, intestinal barrier function and immune training development.

Physician Wu Dongwen is currently a postdoctoral candidate co-supervised by Prof. Liu Xiaowei of Department of Gastroenterology of Xiangya Hospital and Prof.Mandy J McGeachy of the University of Pittsburgh. The team of Prof. Liu Xiaowei has been long devoted to the clinical, basic, and translational research on intestinal barrier, enterohepatic circulation and autoimmune digestive disorders. With over 20 SCI papers published on international journals such asGastroenterology,Cell Death & Differentiation,Inflammatory Bowel Diseasein the past five years,he maintains stable research orientations and sophisticated research platforms.
